  • PeptiSleep™: the innovative compound PeptiSleep™ is a hydrolysate of brown rice (Oryza sativa) proteins. Its bioactive, short, plant-derived peptides have been discovered using artificial intelligence. The time taken to fall asleep was reduced, night-time awakenings were reduced and participants reported more restful sleep without drowsiness the following day. These effects were consistent in terms of both subjective criteria (PSQI, self-observation diaries) and objective criteria (actigraphy), validating the potential effectiveness of PeptiSleep™ as a natural peptide-based sleep aid. In particular, the study reports these figures: +32% Heart Rate Variability (an indicator of the body's ability to recover), +12 min of REM sleep, +13 min of deep sleep and +35 min of total sleep on average (1);
  • L-ornithine: L-ornithine is an amino acid produced naturally by the liver during the urea cycle, a process that eliminates ammonia from the body. L-ornithine supplementation is being studied for its potential ability to reduce levels of cortisol ('the stress hormone') and limit its nocturnal peaks, thereby reducing nervousness and anxiety and their impact on sleep quality. An animal study suggests that L-ornithine also owes its relaxing properties to its regulatory action on the brain's GABA-A receptors (2). L-ornithine also appears to increase the secretion of hormones released during deep, slow-wave sleep, which may indicate its contribution to maintaining the N3 phase (3). Another study has shown that orally administered L-ornithine reaches the brains of mice and, at moderate doses, lowers their stress hormone levels via a mechanism independent of the main brain neurotransmitters (4). In a randomised controlled trial in 2014 involving 52 people, taking 400 mg of ornithine at bedtime for 8 weeks significantly reduced levels of stress hormones and improved sleep quality (subjective scores on insomnia scales) compared with a placebo (5). Finally, a randomised double-blind study involving 22 members of a Japanese research expedition to Antarctica also proved enlightening. During their 4-week stay, part of the team took L-ornithine (400 mg/day). Although the sleep of all the members deteriorated in the difficult conditions of Antarctica, taking L-ornithine seems to have led to a clear improvement in the test group compared with the placebo group (6);
  • an extract of Magnolia officinalis bark containing 90% honokiol and magnolol: magnolia is a tree native to Asia and North America, recognisable by its large pink or white flowers. Its bark contains honokiol and magnolol, phenolic compounds that have been studied for their potential ability to act on GABAergic receptors in the brain, thereby reinforcing the calming effect of GABA. These effects could make it easier to fall asleep and reduce anxiety. The active ingredients in magnolia could also help to modulate the HPA (hypothalamic-pituitary-adrenal) axis, involved in regulating cortisol, whose nocturnal elevations are known to fragment sleep. As animal studies have shown, honokiol promotes non-REM sleep, including deep phases, while preserving the slow wave (delta) activity characteristic of truly restorative rest (7). Human clinical studies on the reduction of cortisol and stress have evaluated formulations including magnolia, with excellent results (8);
  • L-theanine: L-theanine is an amino acid found in tea leaves, reputed to promote relaxation and good sleep patterns. This active ingredient seems to cross the blood-brain barrier easily and modulate neurotransmitters in the brain. Specifically, it inhibits the release of glutamate (the main excitatory neurotransmitter) and increases levels of GABA (the main calming neurotransmitter) and dopamine. L-theanine is thus thought to increase the alpha waves in the brain, associated with relaxation. It is thought to reduce hyperarousal (excessive brain activity), favouring a state of relaxed wakefulness conducive to rest, and to help maintain deep N3 sleep without causing sedation or impairment of faculties the following day. Several clinical studies suggest that L-theanine supplementation can reduce stress and anxiety. In a 2019 double-blind randomised placebo-controlled crossover trial (involving 30 healthy adults taking 200 mg/day of L-theanine for 4 weeks), participants reportedly showed a significant reduction in anxiety-trait (STAI-T) and stress symptoms, as well as an improvement in sleep quality compared with the placebo. Sleep duration and sleep disturbance scores, assessed by a standardised questionnaire (PSQI), also improved with L-theanine compared with the placebo (9). A more recent randomised controlled trial from 2024 (involving 30 adults suffering from moderate stress, taking 400 mg/day for 28 days) indicates that L-theanine significantly reduces perceived stress (PSS score dropping by 18% at 4 weeks) and supports uninterrupted, quality sleep, with good tolerability(10). Another trial on acute stress (randomised, triple-blind crossover), using a single dose of 200 mg, appears to confirm that L-theanine increases frontal alpha brain waves and produces a sharp drop in salivary cortisol (11).

How can I stop waking up at night?

There are a number of good habits you can adopt to reduce the number of night-time awakenings:

  • manage your stress better, particularly at the end of the day (breathing, relaxation, calm activities, etc.)
  • reduce your consumption of stimulants, such as caffeine, at the end of the day
  • adopt regular bedtime and wake-up times
  • avoid exposure to screens in the hour before bedtime
