Affron® saffron extract: saffron is a spice obtained by drying the red stigmas of the crocus flower (Crocus sativus). It contributes to emotional balance, helping you to relax and be in a good mood by naturally stimulating the production of neurotransmitters such as serotonin and dopamine. Affron® saffron extract is standardised with over 3.5% Lepticrosalides®, a patented full-spectrum blend of synergistic bioactive compounds responsible for saffron's effectiveness against depression, anxiety and insomnia, according to 9 clinical studies. This saffron extract is also the most clinically studied (1-11). One randomised, double-blind, placebo-controlled study (N = 128) showed that taking Affron® (28 mg/day for 4 weeks) significantly reduced mood disorders such as tension, anger, depression, fatigue and confusion (12). Another randomised clinical trial found that taking Affron® (28 mg/day for 28 days) improved the quality and restorative nature of sleep, with fewer night-time awakenings and increased wakefulness, with the main benefits appearing as early as the first week (13). In each case, Affron® was well tolerated, and no side effects were reported. Magnesium bisglycinate: magnesium is a mineral which contributes to the normal functioning of the nervous system by regulating neuronal excitability. It also helps to reduce fatigue by activating ATP, the cellular energy molecule. Finally, it contributes to normal psychological functioning by promoting the activity of GABA (an inhibitory neurotransmitter) and reducing the release of stress hormones such as adrenaline and cortisol. If low magnesium levels increase sensitivity to stress, excess stress in turn leads to increased mobilisation and elimination of magnesium, maintaining a 'vicious circle' in which magnesium deficiency and stress reinforce each other. Five of the 7 studies analysed in a systematic review of clinical trials in 2024 reported a significant reduction in anxiety symptoms thanks to magnesium supplementation (14). In this case, we opted for magnesium bisglycinate, a combination of magnesium and two glycine molecules, which is the most bioavailable form and the best tolerated from a digestive point of view;

L-theanine: this amino acid derived from green tea is mainly responsible for its soothing and relaxing effect, without sedation. L-theanine crosses the blood-brain barrier and appears to compete with glutamate at NMDA and AMPA receptors, thereby attenuating excitatory signalling. L-theanine is also thought to reduce glutamate release while increasing GABA levels and promoting the release of glycine, dopamine and serotonin in the brain. Finally, it is thought to increase alpha wave activity, a pattern associated with relaxed alertness. In a double-blind randomised crossover clinical trial in 2019 (30 healthy adults, 200 mg/day of L-theanine for 4 weeks), participants showed a significant reduction in anxiety and stress-related symptoms, as well as an improvement in sleep quality compared with placebo (15). Another trial on acute stress using a single dose of 200 mg (randomised, triple-blind crossover) showed that L-theanine increased frontal alpha brain waves and produced a greater drop in salivary cortisol 1 hour after a stressor, compared with placebo (16). These changes correspond to a calming and anxiolytic effect on the brain;

Bioactive vitamin B6 (pyridoxal-5-phosphate): vitamin B6 helps to reduce fatigue by participating in the production of energy from nutrients. It also helps to support the normal functioning of the nervous system and normal psychological functioning by being necessary for the synthesis of essential neurotransmitters (serotonin, dopamine, GABA), low levels of which aggravate sensitivity to stress. It has also been studied to promote magnesium absorption by facilitating its entry into cells. A vast 8-week randomised controlled study (on 264 healthy adults with high stress levels and low magnesium levels) compared the administration of magnesium alone with that of magnesium + vitamin B6. Both groups showed a significant reduction in stress and anxiety scores, from 'severe' at the start of the study to near-normal levels by the 8th week, with significant results as early as the first 4 weeks. The group receiving the Mg + B6 combination showed a greater reduction in perceived stress (17). What's more, the form chosen in our formula, pyridoxal-5-phosphate (P-5-P), is the bioactive form of B6 that can be used directly by the body.
